- Review and Approval of BPRs and Amendments
R24-03-007NS, Amendment 12
Title: Core Non-Infectious BPR
RS: Miranda Nebane
IBC reviewed, comments and contingencies were received. A motion was made to approve
with additions, a second was made, and by voice vote the BPR amendment was approved.
- Agent Name: human FGFR4 and PLC-γ cDNA Inserts
- Agent Characteristics: No change from core
- Containment Conditions: BSL-2
- Sources of the Inserted DNA Sequences: commercially supplied
- Foreign Gene Expression: Hek293t cells express fusion protein
- Protein Expressed: fusion proteins
- Applicable Section(s) of the NIH Guidelines: Section III D 3
- Types of Manipulations Planned: Same as Core
Contingency 1: Section 5.10 – Will cells be fixed before running through the cytometer?
Response:
No, the cells will not be fixed. After the cells are treated with library compounds and infected
with HIV-1, the plates will be sealed and read on the cytometer.
Contingency 2: Section 5.11 – Should be “Section III-D-3-a”
Response:
This has been corrected; thank you.

Title: High Throughput Screening of Broad-Spectrum Viruses at BSL2 and BSL3
RS: Miranda Nebane
IBC reviewed, comments and contingencies were received. A motion was made to approve
with additions, a second was made, and by voice vote the BPR was approved. - Agent Name: Various Viruses (including Influenza, SARS-CoV, SARS-CoV-2, MERS
CoV, ZIKA, Dengue, HIV-1, EEE, WEE, CHIKV, RSV, Vaccinia, Monkeypox - Agent Characteristics: Various (depending on the virus)
- Containment Conditions: BSL-2 / BSL-3
- Sources of the Inserted DNA Sequences: Commercial and collaborators
- Foreign Gene Expression: Various, such as GFP reporter and NanoLuc reporter
- Protein Expressed: Various, such as GFP and NanoLuc
- Applicable Section(s) of the NIH Guidelines: Section III-D
- Types of Manipulations Planned: No recombinant manipulation at SR
Contingency 1: Section 5.1 – * denotes BSL-3 requirements. SARS-CoV-2 is no longer
classified as BSL-3. See line c. in the listed viruses. It is also listed again in line p. Is this a
duplicate?
Response:
These have been corrected. Thank you for catching them.

Contingency 2: Section 5.8 – “Other” is checked, but there is no material listed.
Response:
This has been unchecked and 5.8(c) modified to better explain.
Contingency 3: Should BSL-3 be checked in section 5.10b?
Response:
Yes, BSL-3 should absolutely be checked. I have corrected that; thank you.

Title: Toxicity and Biodistribution Study of EVO-001 Test Material following a Single
Intravenous Dose Administration with 6-Weeks of Recovery in Cynomolgus
Macaques
RS: Erin Luea
IBC reviewed, comments and contingencies were received. A motion was made to approve
with additions, a second was made, and by voice vote the BPR was approved. - Agent Name: EVO-001
- Agent Characteristics: a single-stranded non-replicating recombinant AAVMYO2
vector - Containment Conditions: BSL-2, A/BSL-2
- Sources of the Inserted DNA Sequences: Supplied by study sponsor
- Foreign Gene Expression: n/a
- Protein Expressed: n/a
- Applicable Section(s) of the NIH Guidelines: III-D-3-a, III-D-4-b
- Types of Manipulations Planned: AAV negative NHPs will be given an
immunosuppressant to and dosed with EVO-001. Blood, urine, and tissue samples will
be collected to determine the toxicity and biodistribution of EVO-001.
Contingency 1: Section 4.2 – I cannot locate this specific training and do not recall it (but
could have missed it). Can safety confirm it is available? Training for this specific type of
NHP (AAV Negative) is not completed. An SOP has been submitted into Veeva, but it is not
released.
Response: I am not sure that there is specific NHP AAV Training, however, I know there
was AAV training at one time. We can request this be sent out to all applicable staff again.
Contingency 2: Section 5.3 – Sani-Plex is used in the A/BSL2 – Can this be confirmed if it
will be an approved disinfectant?
Response: Sani-Plex was added as an ‘other’ and I did confirm that it would be appropriate
for use on NHP studies.
Contingency 3: Section 5.3(i) – Please check Coverage+ NPD for ABSL-1 &2 CCP for use
at necropsy.
Response: Coverage + NPD has been marked.
Contingency 4: Section 5.9 – List appropriate PPE for AAV Negative animals (N95 without
an exhaust, Tyveks only, no PAPR, etc) – This states animals are being chair trained but
chair training is a stressful event and has not been approved.
INSTITUTIONAL BIOSAFETY COMMITTEE
Minutes, Page 4 of 4
Response: The list of applicable PPE was included. Designation for chair training has been
removed due to limitation of time for appropriate training.
Contingency 5: Section 5.10: Section (e) is not completed.
Response: Section 5.10(e) has been updated to include the standard PPE and use of BSC.
Contingency 6: Section 5.11 – Add SOP-00555 (General Guidelines for Handling AAV Non
Human Primates).
Response: This SOP has been added to 5.11 (b) however, the SOP does not seem to be
released in Veeva at this time.
Contingency 7: Section 5.11(d) – Specify that only N95s without exhaust are allowed. Lab
coats are not allowed and PAPRs are not allowed for animal safety – not TA safety.
Response: Section was updated to align with Section 5.9 (g).
Contingency 8: Section 5.11(e) is not answered.
Response: Section completed.
Contingency 9: Section 5.13 – Check BSL-2 to cover In Vitro activities.
Response: This has been designated.
